In vitro studyHistamine inhibits the production of ROS through the type 2 Histamine receptor (H2 receptor). Histamine inhibits the production and release of reactive oxygen species (ROS) in monocytes/macrophages (MO) during burst respiration. Histamine and interleukin-2 (IL-2) act synergistically to activate the cytotoxicity of NK cells (NKCC). Histamine in combination with IL-2 can improve response rates and disease-free survival by protecting cells of the immune system from damage by oxidative stress and inducing innate endogenous immune cell toxicity.
In vivo studyIn a rat model of early alcoholic liver injury, Histamine treatment (0.5 mg/kg or 5.0 mg/kg twice daily) prevented liver injury, normalized serum transaminase levels, and significantly reduce the proportion of liver pathology. The protective effect of Histamine was blocked by ranitidine (10 mg/kg) and H2 receptor antagonists, suggesting that histamine effects are mediated primarily through H2 receptors. In the nucleus accumbens of male rats, Histamine (30 picograms/rat, ICV) increased both the accumulation of 3,4-dihydroxyphenylalanine and the concentration of 3,4-dihydroxyphenylalanine, and the effect was not affected by the H2 antagonist zolantidine, suggesting that histamine stimulates brain limbic DA neurons through its action on H1 receptors. Histamine (0.5 mg/kg S.C.) reduced liver tumor weight by 46% and subcutaneous tumor weight by 41% compared to rats injected subcutaneously with normal saline. The antitumor effect of subcutaneous histamine injection was inhibited by ranitidine (50 mg/kg, subcutaneously) in rat malignancies. In Sprague-Dawley rats, Histamine (1000 mg/kg, S.C.) showed acute tissue damage after 24 h and pathological inflammation at the injection site on days 5 and 28. In male Sprague-Dawley rats, Histamine (1000 mg/kg, subcutaneously) resulted in a C max of 167 mM, a t max of 0.5 H, a t 1/2 of 0.95,AUC is 186 mmol-h/L.
